I came off Effexor for three weeks and then went back on due to anxiety.
Should it work right away again?
I do no feel like I did before, still anxious etc.
Was off it 3 weeks and have been back on it three days.

During those three weeks your receptors normalized. You will have to wait the usual length of time it took to work the first time to some extent, it may "kick in" somewhat faster, but have patience.

Withdrawal symptoms should be alleviated immediately. Your symptoms were not consistent with withdrawal, however, but a relapse of anxiety. These symptoms will reduce more gradually over the next few weeks.
The fact that your symptoms did not immediately disappear is further evidence that they were not withdrawal symptoms. This is useful information to know.
I think you will notice clear improvement after a couple of weeks, which will then increase until you feel like you did before. It may of course take some time to fully get over the psychological trauma of your relapse.
