Tired and sleepy on Moclobemide (Manerex)

Posted by adamie on November 1, 2001, at 12:42:38

hi. I have been on Moclobemide for my 3rd day now (also on zyprexa for 2 weeks or so). All AD's I've tried before this have made my depression worse. I cant tell yet if that is happening with the Moclobemide. But I am much more tired and sleepy than usual. Should I take my two doses of 300mg in the night time? I currently take my first dose around 12:30pm and the second around 8pm. I am the most tired around 4pm. I take my zyprexa 5mg around 10pm. Would it be okay to take a moclobemide dose along with the zyprexa? And then perhaps take my second dose at 11:30pm before going to sleep. Or maybe at 9pm and then at 11:30pm.

This may be relavent, it is from looking up
some quick information on drug interactions.

Both Manerix and Zyprexa are metabolized by
cyp450-1a2. I believe that both meds can have serious interactions
with drugs that inhibit cyp450-1a2 like cimitidine.
Zyprexa is also metabolized by cyp450-2d6, which
manerix may inhibit (according to a chart I have),
but I wouldn't venture to guess the net sum
of interactions.

The drug interactions between Manerix and these
newer meds are not well studied. Could be they
are competing for the same enzyme substrate and
this is increasing the zyprexa in your body.
I dunno.

I found that Manerix had an anti-anxiety affect,
but it didn't make me tired. It didn't seem to
matter much for me when I dosed it either. But
this is me, and Manerix was the only med I was
on.

I was on Manerix for 3 years and it didn't make me tired either. I was on 600mg a day and it didn't matter when I took it.
Must be an interaction thing. I hope it works out for you. I did find that it took about 8 weeks to get any benefit from the moclobemide.

Hi Adamie,
You could try taking Zyprexa earlier in the evening. That will lessen the effects the next day. Since you get most tired about 4 hours after Moclobemide dosing, then maybe take the dose 4 hours before bedtime. Take the other dose and cut it into three pieces. Take the pieces in three divided doses throughout a 24 hour period.

Since Moclobemide is increasing serotonin levels, that could also have something to do with the tiredness. If that is the case, then that is a side effect that should lessen and go away in time.

The possibility exists that Moclobemide is increasing the blood levels of Zyprexa. If that is the case, then your 5mg dose of Zyprexa could be more than it appears.

In any case, the dosing strategy I mentioned above could be helpful.
John

I think I'll be stopping my zyprexa. I was on it before and it didn't seem to help. it just simply didn't make my depression worse I felt. i'll see how i do on just the moclobemide. if i continue to be tired then i'll take it near bedtime.

things starting to look scary

yesterday I didn't take my 5mg dose of zyprexa. Today I will still pretty tired but not enough for me to go and fall asleep in the afternoon. I have been feeling as if my mood was pretty much the same (hard to tell) but now things are starting to look scary. I could look at anything and things appear slightly frightening. I think I will keep on taking my zyprexa and just take the moclobemide all in the evening. I dunno what the heck to do.

actually I think I will stick with the moclobemide and see how it goes. it would be a shame if the zyprexa was really helping despite feeling pretty horrible on it, but not suicidal. i'll see how it goes. clearly i cant take both meds since they interact with eachother.

It has to be the combination. Manerix causes insomnia for most people.
It does make me feel tired, but only on a high dose. 900mg or higher. In lower dosage, it's been quite anxiogenic for me. Not activating in a good way, but pure anxiogenic! It's strange to hear that it relieves anxiety in some cases.
