Ok, this is becoming ulmost rediculus. Been trying more & more addons the my MAOI (Marplan) but each trials has its own set of problems. So now Ive just stopped amitriptyline Oi strait on Valdoxan (agomelatine). One of the newwer ADs im told. I certainly like the idea behind it. With the m1 & m2 meletonin receptors agonist effects Im hoping it will correct my sleeping patterns and do away with my heypersomnia. Also is a 5-HT2C antagonist Which in turn boosts NE/na & DA (?). Hope this is the big gun I need...

So I went to sleep by 9pm. Its now 6am and I woke 2hrs ago. WOW! Normally I'd be hanging but I feel wide awake. Start of something new?
And any Valdoxan users out there, past or present please share your experiences.med

Yes. I believe that this is possible. I took my first dose of Valdoxan at bedtime. I woke up feeling significantly better. No placebo here. However, this antidepressant response lasted for no longer than three days. I was surprised that it returned two weeks later. Unfortunately, after another three days, I relapsed and quit taking Valdoxan, perhaps prematurely. I have since added Abilify and am receiving significant from it. I am very seriously thinking about re-trialing Valdoxan, now that Abilify is onboard and the terrain of my brain has changed.

Sorry to butt in, sounds like what happened when i tried remeron, it started to work a week and a half later than stopped. 5 HT2C antagonism is great, but I think sometimes you need an increase in serotonin to keep it from pooping out. Perhaps 5 HT1a agonism keeps the dopamine norepinephrine disinhibition going?

Thanks Scott very interesting. Perhapse all it did for you was normalise your sleep and thats all? It is also said that 25mg is NOT actually the reall dose for best theraputic responce, yet many docs stop at 25mg. 50 sounds more like it. Did you go to 50mg?
And personally, if all it does do is normalise my sleep patterns, then thats a big bonus for myself.
I'll keep Abilify in mind too. May I ask, whats you diagnosis(s) and what does the Abilify help with specifically? Thanx.med

Hey, butt in whenever you like! :)
You got me thinking, esp as theres only so much of an assortment of meds my doc will give me-and rightly so. HOWEVER, one drug available OTC is diphenhydramine, and its said to be a SRI. How specific and how potent I dont know, but i might be a possible candidate in your theory that a serotonin enhancer might keep a 5 HT2C antagonist from poopin out. What ya think?

I tried taking Valdoxan at a dosage of 50mg for about a week. I felt somewhat more depressed. This is a point of interest to me as melatonin supplements make me feel the same way.

Abilify is very capable of producing increases in mental energy and motivation. However, in my experience, I found that it may not improve anhedonia as much. Abilify helps me more when I am taking other medications at the same time.
Currently:
Nardil 90mg
nortriptyline 150mg
Lamictal 200mg
Abilify 10mgI see that many people with unipolar depression do well at 2.5 to 5.0 mg. of Abilify. I actually do best at 10 mg. 20mg made me feel somewhat muddy.

Hang on hang on. Why am I thinking of diphehydramine, when I have plenty of amnitryptyline left over? Im sure 25 to 50mg would suffice to create the SRI conditions on which your theory is based.
I also still have plenty of trimipramine left. It has much different & more actions inc 5-HT2 antagonism. Lots of other effcts tho http://en.wikipedia.org/wiki/Trimipramine
Im not going to play with all that at once- Im actually trying to get better lol. But its nice to know what ur arsnel is ;) lol

An example of the important of SRI with 5 HT2 antagonism is Serzone. It is a potent 5 HT2 receptor antagonist even at low doses, but it is not until you get to the higher doses where there is more SRI that it starts to become an effective antidepressant. I don't know what you should add to help with the serotonin, or if it would definitely work. I would talk to your doctor, I don't know anything about what interacts with valdoxan.

So far so good- well no negatives. Though I have completely lost my appetite. Feels just like my 1st week on buspirone. I'm on a diet of whole milk & fruit, its all I can manage. ATM I dont consider this a negative. I need to loose a little weight. As long as it doesn't get worse I deal with it. Iteresting tho... Any ideas wich mechanism might be causing loss of appetite? Or maybe its a rebound from amitryptyline? Tho I had no particular apetite on that.

Anyway, One thing bizzare which happened yesterday is that after a few cannabis joints I was getting closed-eye hallucinations. I NEVER get them with cannabis alone. Melotonin playing a part? Hhmmm...

Since last Sunday, 5 days. Yet I never had appetite issues whilst i was on it. So perhapse this is withdrawal more than anythings elso, oh and im stil on liquids.

The improved sleep from agomelatine is gone with me, but I stll take it because of the push it gives me.
I cope much better with crappy sleep the day after with it.

Ive already tried adding trimipramine, a particularly strong 5-HT2 agonist to agomelatine. Too many dreams for me in one night thankyou! But just showing it can be done. No toxic 'interaction' from what I felt. But I easily have bad/vivid dreams. It might be an excellent 5-HT2 agonist combo for some...
